Evaluation of Extracellular Circulating Human MicroRNA-197 as a Target Biomarker in Patients with Coronary Artery Disease
نویسنده
چکیده مقاله:
Background: Coronary Artery Disease (CAD) refers to the reduction or blockage of all or part of the coronary arteries due to the process of atherosclerosis or the presence of a clot. The aim of this study was to investigate the association of serum miR-197 as a diagnostic index in patients with coronary artery disease. Methods: In this study, 100 patients with CAD were selected. Extraction of microRNAs from peripheral blood plasma was performed using an exclusive microRNA extraction kit. Then the cDNA synthesis and the expression level of miR-197 were performed using the Real Time RT-PCR technique and the SYBER Green method, using U6 snRNA as an internal control. T-test was used to analyze microRNA expression and significance and to determine the relationship between microRNA expression levels, Pearson correlation coefficient test was used. Results:The results showed a positive correlation between miR-197 expression and CAD. The average expression of 0.78 in the control sample was increased to 2.76 according to severity of involvement in the patient. That is, the relative expression of miR-197 in the patients + group was significantly increased compared to control (P <0.004). Conclusion: It appears that miR-197 can be considered as an indicator of coronary endothelial cell function. It is possible to use it as a bio marker for prognosis, and control of treatment for CAD.

Evaluation of iron status in patients with coronary artery disease
Background: Iron can causes lipid peroxidation in vivo and in vitro. Iron promotes myocardial injury in experimental animals. Low Density Lipoprotein (LDL) oxidation by oxidants including iron have a central role in atherosclerotic process. By this means, evaluation of serum iron in patients with coronary artery disease (CAD) has a significant importance. Materials and Methods: In this cross s...
متن کاملRole of microRNA as a biomarker in Alzheimer’s disease
Introduction: MicroRNAs are small, non-coding, and protected RNA molecules that regulate gene expression after transcription by mRNA degradation or inhibition of protein synthesis. The function of these molecules is critical to many cellular processes, including growth, development, differentiation, homeostasis, apoptosis, aging, stress resistance. In addition, some diseases including cancer, a...
متن کاملCirculating microRNA-126 in patients with coronary artery disease: correlation with LDL cholesterol
BACKGROUND Coronary artery disease (CAD) is a major problem worldwide. Atherosclerosis and thrombosis underlying CAD involve multiple cell types. New and useful diagnostic markers are required. MicroRNAs (miRNAs) are a class of noncoding RNAs that posttranscriptionally regulate the gene expressions involved in various cellular processes. Endothelial dysfunction is implicated in early processes ...
متن کاملMicroRNA Expression in Circulating Microvesicles Predicts Cardiovascular Events in Patients With Coronary Artery Disease
BACKGROUND Circulating microRNAs (miRNAs) are differentially regulated and selectively packaged in microvesicles (MVs). We evaluated whether circulating vascular and endothelial miRNAs in patients with stable coronary artery disease have prognostic value for the occurrence of cardiovascular (CV) events. METHODS AND RESULTS Ten miRNAs involved in the regulation of vascular performance-miR-126,...
